Simultaneous Expression of Bacillus licheniformis Maltogenic Amylase and Ther - mostable Amylase in Recombinant Escherichia coli

Abstract
Characterization of the physiological properties of a recombinant Escherichia coli HBl01/pTMA322 strain which simultaneously produces Bacillus licheniforrnis maltogenic amylase (BLMA) and thermostable amylase (BLTA) was undertaken. Specific growth and substrate consumption rates of the simultaneous expression system were lower than the corresponding values of the single expression systems which produce either BLMA or BLTA. However, the simultaneous expression system was more efficient in product yields compared with the single-expression systems and a mixed culture of the two single-expression systems. Among the amino acids tested, the addition of glycine was most effective in improving cell growth and enzyme production. Supplementation of glycine increased BLMA activity by twofold and BLTA activity by 1.5-fold compared with the control cultivation in the minimal medium.
